Deep-mining Antibody Repertoires of Transgenic Mouse Models with High-throughput Single Cell BCR & WT Sequencing

Overview

Here, we describe a split-pool combinatorial barcoding approach to enable high-throughput single cell BCR and whole transcriptome profiling from fixed human, mouse, or transgenic mouse samples. This method allows for profiling of up to 1M cells or 96 samples at a time resulting in whole transcriptome and BCR libraries. BCR libraries are generated through hybridization based enrichment which allows for versatility across transgenic mouse strains without the need for custom primer design. Using this approach we capture full length paired heavy and light chain sequences with high sensitivity. This method paves the way for high-throughput screening of antibody repertoires from all B cell subtypes.
